Tau targeting RNA aptamers are single-stranded oligonucleotides specifically designed to bind to the human tau protein, a microtubule-associated protein implicated in neurodegenerative diseases known as tauopathies, including Alzheimer's disease. These RNA aptamers inhibit the oligomerization of tau, which is considered a critical step in the formation of toxic aggregates associated with neuronal dysfunction and cell death. By binding to tau and preventing its assembly into oligomers, these aptamers have demonstrated neuroprotective effects in vitro and in cellular models by reducing proteotoxic stress, alleviating synthetic tau oligomer-mediated neurotoxicity, and preserving dendritic spine density. The approach represents a targeted strategy for modulating pathogenic processes underlying Alzheimer's disease and related disorders[2][1][5].
