TAVO101 × IL4R-dupi is a bispecific antibody (BsAb) designed to simultaneously target thymic stromal lymphopoietin (TSLP) and the interleukin-4 receptor alpha (IL-4Rα). Developed by Tavotek Biotherapeutics, the molecule is constructed using controlled Fab-arm exchange, combining the Fab arms of the anti-TSLP antibody TAVO101 and the anti-IL-4Rα antibody dupilumab. TSLP acts as an upstream epithelial alarmin that initiates type 2 inflammation, while IL-4Rα is a shared receptor component for the downstream effector cytokines IL-4 and IL-13. By dual-targeting these pathways, the drug aims to provide more comprehensive control of allergic and chronic inflammatory diseases, such as asthma and atopic dermatitis, compared to single-target blockade. The antibody features an engineered IgG1 Fc region containing AALS mutations (L234A, L235A, M428L, and N434S) intended to extend its half-life through enhanced neonatal Fc receptor (FcRn) binding and to minimize effector functions like antibody-dependent cellular cytotoxicity (ADCC).
