TAVO423 is a trispecific antibody-enzyme (AbEn) conjugate designed to modulate the tumor microenvironment (TME) by targeting cancer-associated fibroblasts (CAFs) and degrading the extracellular matrix component hyaluronan (HA). The molecule consists of a trispecific antibody framework that binds to Fibroblast Activation Protein (FAP) and Leucine-Rich Repeat Containing 15 (LRRC15), fused with a recombinant human hyaluronidase (HYAL) enzyme. By dual-targeting FAP and LRRC15, TAVO423 achieves localized delivery of hyaluronidase to the stroma of solid tumors. The subsequent enzymatic depletion of HA reduces interstitial fluid pressure and removes physical barriers, thereby enhancing the intratumoral penetration of co-administered therapies such as chemotherapy and immune checkpoint inhibitors, while also promoting the infiltration of cytotoxic T lymphocytes. Developed by Tavotek Biotherapeutics, TAVO423 has demonstrated preclinical efficacy in gastrointestinal cancers, including colorectal and pancreatic models, as well as breast cancer.
