TAVO605 is a novel antibody-drug conjugate (ADC) currently in preclinical development by Tuochuang Biotechnology (also known as Tavo Biopharma). It targets CD318, also known as CUB Domain Containing Protein 1 (CDCP1), a single-span transmembrane receptor that is significantly upregulated in several major solid tumors—including breast, lung, pancreatic, colorectal, bladder, and prostate cancers—and is associated with poor prognosis. The drug conjugates an anti-CD318 antibody with the cytotoxic payload monomethyl auristatin E (MMAE), a tubulin inhibitor, at a drug-to-antibody ratio (DAR) of 4. In preclinical studies, TAVO605 demonstrated potent cytotoxicity across multiple cancer cell lines and showed strong tumor growth inhibition in CDX and PDX xenograft models of pancreatic, colon, triple-negative breast, and non-small cell lung cancers. It is being developed as a potential therapy for solid tumors that are non-responding or resistant to Enhertu (trastuzumab deruxtecan) treatment.
