This is a combination regimen consisting of four agents: - **Tavocept** is a chemoprotective agent developed to reduce the toxicity of platinum-based chemotherapy regimens such as those containing cisplatin. Its primary mechanism involves reducing nephrotoxicity and other side effects associated with platinum drugs[7]. - **Cisplatin** is a platinum-based chemotherapeutic that forms DNA crosslinks and inhibits DNA synthesis and function. - **Docetaxel** is a semisynthetic taxane that binds to the β-subunit of tubulin in microtubules. It promotes microtubule assembly while inhibiting disassembly, leading to stabilization of microtubules and inhibition of cell division. This results in mitotic arrest and apoptosis[2][6][8]. - **Paclitaxel** is also a taxane that stabilizes microtubules by binding to the β-subunit of tubulin at the same site as docetaxel but with lower affinity. Both taxanes disrupt normal mitotic spindle formation. This combination has been studied primarily for use in advanced non-small cell lung cancer (NSCLC) where standard chemotherapy regimens include either docetaxel or paclitaxel plus cisplatin; tavocept may be added for its protective effects against toxicity[1][7].
