Overview

Tavokinogene telseplasmid is a synthetic DNA plasmid encoding the human interleukin 12 (hIL‑12) protein, specifically both the p35 and p40 subunits, separated by an internal ribosome entry site and under control of a cytomegalovirus promoter. It is delivered via intratumoral injection followed by electroporation to enable local expression of IL‑12 in the tumor microenvironment. The expressed IL‑12 acts as an immunostimulant, enhancing anti-tumor immune responses by promoting activation of cytotoxic T cells and natural killer cells. Tavokinogene telseplasmid is being developed primarily for various cancers including malignant melanoma, Merkel cell carcinoma, cutaneous T-cell lymphoma, head and neck cancer, and triple negative breast cancer. The drug has orphan designation for malignant melanoma[1][2][3][5][6].
