This is a multi-agent combination regimen that combines four agents: - **Tavokinogene telseplasmid** (OBT-206), an investigational DNA-based gene therapy designed to express interleukin-12 (IL-12), thereby enhancing antitumor immunity. - **Pembrolizumab**, a monoclonal antibody against programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance T-cell anti-tumor activity. - **Gemcitabine** and **Carboplatin**, both standard cytotoxic chemotherapeutic agents. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, while carboplatin is a platinum-based DNA crosslinker, inducing apoptosis in rapidly dividing tumor cells. This four-drug combination is under evaluation for synergistic immunostimulatory effects and cytotoxicity in cancers, most notably in solid tumors such as bladder cancer and possibly other advanced malignancies.
