Overview

Taxane + capecitabine is a combination chemotherapy regimen used primarily in the treatment of breast cancer, especially metastatic and locally advanced cases. Taxanes (such as docetaxel or paclitaxel) are microtubule inhibitors that disrupt cell division by stabilizing microtubules and preventing their depolymerization. Capecitabine is an oral prodrug that is converted to 5-fluorouracil (5-FU) preferentially within tumor cells via thymidine phosphorylase upregulation—a process enhanced by prior exposure to taxanes—leading to inhibition of DNA synthesis and RNA function. The combination exploits this synergy for improved antitumor activity compared to either agent alone. This regimen has demonstrated higher response rates and progression-free survival in metastatic breast cancer compared with single-agent therapy, though toxicity profiles differ depending on scheduling[1][2][8]. Capecitabine is marketed under the brand name Xeloda.
