Overview

This is a combination regimen consisting of a taxane (such as docetaxel or paclitaxel), trastuzumab, and pertuzumab. It is primarily used as first-line therapy for HER2-positive breast cancer, including both early-stage and metastatic settings. Taxanes are antimicrotubule agents that inhibit cell division by stabilizing microtubules, leading to cancer cell death. Trastuzumab and pertuzumab are monoclonal antibodies targeting the human epidermal growth factor receptor 2 (HER2). Trastuzumab binds to subdomain IV of HER2, while pertuzumab binds to subdomain II; together they provide dual blockade of HER2 signaling pathways, resulting in synergistic antitumor activity. This combination has demonstrated improved progression-free survival and overall survival compared to regimens without dual anti-HER2 therapy[1][3][4].
