Overview

**Tazemetostat + itraconazole** is a combination of two small molecule drugs: tazemetostat, a histone-lysine N-methyltransferase EZH2 inhibitor approved for epithelioid sarcoma and certain follicular lymphomas, and itraconazole, an oral triazole antifungal agent primarily used for systemic fungal infections. Coadministration of itraconazole (a strong CYP3A4 inhibitor) with tazemetostat results in increased tazemetostat plasma exposure due to reduced metabolism of tazemetostat, which could impact both efficacy and safety. This combination is not an approved fixed-dose co-formulation, but the drug-drug interaction is clinically relevant in oncology and infectious diseases[1][2][3][4].
