Overview

TB-003B is a preclinical-stage proteolysis-targeting chimera (PROTAC) designed to induce the targeted degradation of cyclin-dependent kinase 9 (CDK9). Developed by Therabene, it serves as a backup molecule to their lead candidate, TB-008. CDK9 is a key component of the positive transcription elongation factor b (P-TEFb) complex, which regulates the transcription of short-lived proteins such as c-Myc and MCL-1 that are often dysregulated in various malignancies. By utilizing the PROTAC modality, TB-003B aims to achieve more potent and sustained inhibition of CDK9 signaling compared to traditional small-molecule inhibitors. The program is being explored for potential applications in oncology, autoimmune disorders, and viral diseases.
