Overview

TB001 is a dual glucagon-like peptide-1 receptor/glucagon receptor (GLP-1R/GCGR) agonist with higher affinity towards GCGR. It has been developed as a potential therapeutic for liver fibrosis, demonstrating remarkable potency, selectivity, extended half-life, and low toxicity in various rodent models. TB001 attenuates liver injury and collagen accumulation in multiple animal models of liver fibrosis, including those induced by CCl4, α-naphthyl-isothiocyanate (ANIT), bile duct ligation (BDL), and Schistosoma japonicum infection. TB001 works by inhibiting the activation of hepatic stellate cells through suppression of TGF-β expression and downstream Smad signaling pathways. It also blocks pro-inflammatory NF-κB/IKBα pathways and reduces JNK-dependent induction of hepatocyte apoptosis.
