TB155D is a preclinical-stage antibody-drug conjugate (ADC) being developed by TwinPigBio for the treatment of cancer. It utilizes the company's proprietary ESCLink™ platform to conjugate a Human epidermal growth factor receptor 2 (HER2)-targeting antibody with a Proteolysis-Targeting Chimera (PROTAC) payload. The therapeutic strategy involves using the antibody to selectively deliver the PROTAC into HER2-positive tumor cells, where the payload then facilitates the ubiquitination and subsequent proteasomal degradation of the cMyc (MYC) transcription factor. cMyc is a prominent oncogenic transcription factor often considered difficult to target with traditional small molecules; by combining the precision of an ADC with the degradative power of a PROTAC, TB155D aims to overcome these challenges in HER2-expressing malignancies.
