Overview

TB206-001 is a fully humanized, high-affinity monoclonal antibody antagonist targeting the adenosine A2A receptor (A2AR), a G protein-coupled receptor (GPCR) implicated in tumor immune evasion. By selectively binding to and antagonizing A2AR, TB206-001 blocks the immunosuppressive effects of adenosine within the tumor microenvironment, thereby restoring immune cell activity and enhancing antitumor responses. Preclinical studies have demonstrated that TB206-001 exhibits potent and selective antagonism of human A2AR with nanomolar affinity, cross-reactivity with mouse and cynomolgus monkey A2AR (but not other adenosine receptors), and robust tumor-suppressing activity in animal models—outperforming existing PD-1 inhibitors. The antibody was discovered using phage display libraries constructed from animals immunized with DNA encoding for A2AR. Twist Bioscience developed this candidate as a potential next-generation cancer immunotherapy checkpoint inhibitor[1][4][5][7].
