TB521

TB521 is a single-target peptide-drug conjugate (PDC) developed by TwinPigBio, designed to target Fibroblast Activation Protein (FAP) on Cancer-Associated Fibroblasts (CAFs). Built on the proprietary TAMpep™ - ESCLink™ platform, it delivers a proapoptotic peptide payload (dKLA) directly to the tumor microenvironment. The dKLA payload (a D-enantiomer of the (KLAKLAK)2 peptide) induces cell death by disrupting mitochondrial membranes upon internalization. By specifically depleting CAFs, which are critical supportive cells in the tumor stroma, TB521 aims to remodel the tumor microenvironment, reduce pro-tumorigenic signaling, and enhance the penetration and efficacy of co-administered anti-cancer therapies. It is currently in preclinical development for the treatment of various cancers.
