Overview

TB571 is a multi-target peptide-drug conjugate (PDC) developed by TwinPigBio using its proprietary TAMpep™ - ESCLink™ platform. It is designed to simultaneously target the tumor microenvironment (TME) by addressing both tumor-associated macrophages (TAMs) and cancer-associated fibroblasts (CAFs). Specifically, the peptide component targets active CD18 on M2-type TAMs and Fibroblast Activation Protein (FAP) on CAFs. The drug delivers a proapoptotic peptide payload, dKLA (D-enantiomer of (KLAKLAK)2), which induces mitochondrial membrane disruption and subsequent apoptosis in these stromal/tumor-supporting cells. By depleting these immunosuppressive and tumor-promoting cells, TB571 aims to inhibit tumor growth and enhance the efficacy of other cancer therapies. It is currently in the preclinical stage of development for various cancers.
