**TB59-2** is a preclinical GLP-1 receptor agonist antibody-derived fusion protein discovered by Twist Biopharma from a synthetic GPCR-focused antibody library. It was generated by fusing a GLP-1 peptide to the light chain of a non-functional GLP-1 receptor-specific antibody, creating an agonistic biologic that activates glucagon-like peptide 1 receptor signaling and stimulates cAMP activity with potency reported to be similar to native GLP-1 7-36 in engineered cell assays. Published preclinical reports describe specific binding to GLP-1 receptor-expressing cells, glucose-lowering activity in animal models, and a longer apparent half-life than free GLP-1 peptide, supporting its development concept for type 2 diabetes and potentially other metabolic indications, although no advanced clinical development has been clearly documented.
