TBA-7371 is an orally active small molecule and non-covalent inhibitor of decaprenylphosphoryl-beta-D-ribose oxidase (DprE1), an essential enzyme in the cell wall biosynthesis pathway of Mycobacterium tuberculosis. By inhibiting DprE1, TBA-7371 blocks the synthesis of arabinose in the bacterial cell wall, leading to structural defects and exerting a potent anti-tuberculosis effect. It belongs to the class of indoles and was identified through lead optimization studies as part of a novel series of 1,4-azaindole antibacterial compounds. The drug has demonstrated efficacy in preclinical rodent models and is currently under clinical evaluation for pulmonary tuberculosis. Its development involves both AstraZeneca and the Global Alliance for TB Drug Development (TB Alliance), with clinical trials also sponsored by Bill & Melinda Gates Medical Research Institute.
