TBC2576 is a **small molecule drug** originally developed by Encysive Pharmaceuticals. It is a potent and highly selective endothelin A (ET_A) receptor antagonist with approximately ten-fold higher binding affinity for ET_A compared to sitaxsentan (TBC11251). The compound has high ET_A/ET_B selectivity and demonstrated in vivo activity with a serum half-life of 7.3 hours in rats. TBC2576 belongs to a chemical series of 2,4,6-trisubstituted anilino ring derivatives optimized for increased potency and selectivity. It was investigated in preclinical and clinical studies for indications such as neoplasms, diabetic neuropathies, glaucoma, and other nervous system or metabolic diseases, but ultimately its development was discontinued[1][3].
