TBI-1201 is an investigational autologous cell therapy composed of human T lymphocytes genetically engineered to express a retroviral vector encoding a high-affinity T-cell receptor (TCR) specific for the melanoma-associated antigen A4 (MAGE-A4). The product is designed to target and kill tumor cells expressing the MAGE-A4 antigen in patients with solid tumors. The approach involves isolating patient-derived (autologous) peripheral blood mononuclear cells (PBMCs), modifying them ex vivo with a retroviral vector carrying the anti-MAGE-A4-TCR gene, expanding these modified cells, and then infusing them back into the patient following preconditioning chemotherapy. The primary indication under investigation has been unresectable or refractory solid tumors that are positive for HLA-A*24:02 and express the MAGE-A4 antigen. Developed by Takara Bio in collaboration with academic partners such as Mie University[1][2][3][5].
