TBI-1501 is an autologous T-cell receptor (TCR) engineered T-cell therapy developed by Takara Bio for the treatment of Wilms' tumor protein 1 (WT1)-positive hematologic malignancies, specifically acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS). The therapy utilizes Takara's proprietary siTCR™ technology, which employs a retroviral vector (pMS3-WT1-siTCR) to introduce a TCR specific for the WT1 235-243 peptide presented by the HLA-A*24:02 molecule. A key innovation of the siTCR platform is the inclusion of small interfering RNA (siRNA) sequences that knockdown the expression of endogenous TCR alpha and beta chains. This reduction in endogenous TCR expression minimizes mispairing between endogenous and exogenous TCR chains, thereby increasing the surface density of the therapeutic TCR and potentially improving both the safety and potency of the engineered T-cells. TBI-1501 is administered via intravenous infusion and has been evaluated in Phase 1 clinical trials in Japan.
