Overview

TBI-302 is a novel protein-drug conjugate consisting of the chemotherapy agent floxuridine covalently attached to hemoglobin. Developed by Therapure Innovations (a division of Therapure Biopharma), this drug leverages the natural clearance pathway of hemoglobin through the liver to selectively deliver cytotoxic therapy directly to hepatic tumors, such as hepatocellular carcinoma (HCC) and other liver cancers. The mechanism of action is inhibition of thymidylate synthase, an enzyme critical for DNA synthesis, via the floxuridine moiety. This targeted approach aims to maximize antitumor activity in the liver while minimizing systemic toxicity typically associated with conventional chemotherapy[1][3][5][9].
