Overview

TBI-New York Medical College-Leukemia-Lymphoma-Bone Marrow Failure refers to a specific therapeutic regimen and cell therapy product developed by New York Medical College for the treatment of high-risk hematologic malignancies and non-malignant blood disorders. The protocol centers on the use of CD34+ selected allogeneic hematopoietic stem cells, which are purified from donor grafts (partially matched family or matched unrelated donors) to reduce the risk of graft-versus-host disease (GVHD) by depleting T-cells. This cell therapy is administered following a Full Intensity with TBI conditioning regimen, which includes fractionated total body irradiation (TBI), the alkylating agents thiotepa and cyclophosphamide, and rabbit antithymocyte globulin (ATG). This combination is designed to achieve complete myeloablation and profound immunosuppression in the recipient, ensuring the successful engraftment of the CD34-enriched donor cells. The regimen is primarily investigated in pediatric and young adult populations for indications such as leukemia, lymphoma, sickle cell disease, and various inborn errors of metabolism.
