Overview

TBID (also referred to as tBid) is a selective small molecule inhibitor of homeodomain-interacting protein kinase-2 (HIPK2). It is primarily utilized as a research tool to investigate the role of HIPK2 in cell proliferation, survival, and disease pathogenesis. In the context of pulmonary arterial hypertension (PAH), TBID has been shown to inhibit the pro-proliferative and pro-survival phenotype of pulmonary arterial smooth muscle cells (PASMCs) by downregulating signaling pathways involving YAP, CEP55, and PKMYT1. Preclinical evidence in rat models of severe pulmonary hypertension indicates that pharmacological inhibition of HIPK2 with TBID can improve systolic right ventricular pressure and attenuate pulmonary vascular remodeling, suggesting its potential as a therapeutic strategy for PAH.
