Overview

TBio-6517 is an engineered oncolytic vaccinia virus designed for the treatment of advanced solid tumors. It is armed with three immune-modulating agents—Flt3 ligand, anti-CTLA-4 antibody, and interleukin 12 (IL-12)—to enhance local immune activation within the tumor microenvironment. The virus selectively infects and lyses cancer cells while delivering these transgenes directly to tumor sites. This multi-modal approach aims to stimulate anti-tumor immunity by promoting dendritic cell recruitment (via Flt3 ligand), blocking CTLA-4-mediated immunosuppression (via anti-CTLA-4 antibody), and enhancing T-cell activation (via IL-12). TBio-6517 is being developed as both a monotherapy and in combination with pembrolizumab for various solid tumors including triple negative breast cancer, microsatellite stable colorectal cancer, melanoma, cholangiocarcinoma, cutaneous squamous cell carcinoma, HPV-positive oropharyngeal squamous cell carcinoma, mesothelioma, renal cell carcinoma, and others[1][2][5][6][7][10].
