Overview

TBRIFISO001 is a fixed-dose combination (FDC) drug candidate developed by Reach Pharmaceuticals for the treatment of tuberculosis. It consists of two first-line antitubercular agents: rifampicin and isoniazid. Rifampicin is a macrocyclic antibiotic that inhibits bacterial DNA-directed RNA polymerase, thereby blocking RNA synthesis. Isoniazid is a prodrug that, upon activation by the catalase-peroxidase enzyme KatG, inhibits the enzyme enoyl-[acyl-carrier-protein] reductase (InhA), which is essential for the synthesis of mycolic acids in the mycobacterial cell wall. This combination is designed to simplify treatment regimens, improve patient adherence, and reduce the risk of developing drug-resistant tuberculosis by ensuring the simultaneous administration of multiple effective agents.
