TBS-2025 is a first-in-class, small molecule dual agonist of the thyroid hormone receptors beta (THR-β) and gamma (THR-γ). Developed by TuHURA Biosciences (formerly Morphogenesis), the drug utilizes the company's proprietary Dual Orally-available Receptor (DOR) technology. TBS-2025 is primarily being developed for the treatment of metabolic dysfunction-associated steatohepatitis (MASH), a condition characterized by liver inflammation and damage caused by a buildup of fat. By selectively activating THR-β and THR-γ, the drug aims to mimic the beneficial metabolic effects of thyroid hormones in the liver—such as reducing hepatic fat and improving cholesterol levels—while avoiding the adverse effects associated with THR-α activation, such as heart rate increases and bone density loss. The dual agonism of both beta and gamma isoforms is intended to provide a more comprehensive therapeutic effect on lipid metabolism and fibrotic pathways compared to single-receptor modulators.
