Overview

TC-2696 is a small molecule selective ligand for the alpha4beta2 (α4β2) nicotinic acetylcholine receptor (nAChR), originally developed by Targacept. It was designed to modulate the cholinergic system to improve cognitive functions, specifically targeting cognitive impairment associated with schizophrenia (CIAS). The α4β2 nAChR subtype is highly expressed in the cerebral cortex and hippocampus, areas vital for memory and attention. Despite showing promise in preclinical models and early-stage safety trials, TC-2696 failed to demonstrate statistically significant improvement over placebo in a Phase 2 clinical trial for CIAS, leading to the discontinuation of its development for this indication.
