TC-510 is an autologous cell therapy consisting of genetically engineered T cells designed to target mesothelin-expressing solid tumors. The therapy incorporates two synthetic constructs: a single-domain antibody that recognizes human mesothelin, fused to the CD3-epsilon subunit and incorporated into the endogenous T cell receptor (TCR) complex, and a PD-1:CD28 chimeric switch receptor expressed independently on the T cell surface. This design aims to enhance anti-tumor activity by both targeting mesothelin-positive cancer cells and overcoming immunosuppressive signals mediated by PD-L1/PD-L2 in the tumor microenvironment. TC-510 was developed as an enhanced version of gavo-cel (TC-210), with improved effector function, resistance to exhaustion, and prolonged persistence of engineered T cells[1][3][6][9]. The drug was developed by TCR2 Therapeutics (a subsidiary of Adaptimmune). As of late 2023, clinical development for TC-510 has been terminated following risk-benefit analysis due to safety concerns and limited efficacy[8].
