Drug intelligence / Profile preview
TCE-09 is a preclinical mRNA-encoded bispecific T-cell engager (TCE) being co-developed by the Dana-Farber Cancer Institute and Nona Biosciences for the treatment of multiple myeloma and other hematological malignancies. The therapeutic candidate utilizes a single-chain mRNA architecture (5'UTR-signal peptide-scFv-scFv-half-life extender-3'UTR) to encode a bispecific T-cell engager targeting B-cell maturation antigen (BCMA) on tumor cells and CD3 epsilon on T cells. By delivering the TCE as an mRNA formulation, the therapy aims to achieve transient, controlled expression of the bispecific molecule in vivo, while the incorporated half-life extender provides prolonged therapeutic activity, offering a potentially safer and more convenient alternative to continuous infusion of recombinant protein-based T-cell engagers.
