TCE-R lipid nanoparticle is a preclinical mRNA-based immunotherapy developed by Abalos Therapeutics, designed to enhance the efficacy of T-cell engagers (TCEs) in treating solid and liquid tumors. The therapy consists of mRNA encoding a T-cell engager receptor (TCE-R) encapsulated in a lipid nanoparticle (LNP) for in vivo delivery. The TCE-R is a universal chimeric adapter protein that, once expressed on the surface of innate immune cells such as macrophages and dendritic cells, allows them to be engaged by any CD3-targeting TCE. The receptor features an extracellular domain recognized by the CD3-binding arm of the TCE and intracellular signaling domains that trigger innate effector functions, including phagocytosis and pro-inflammatory cytokine production. This approach aims to recruit the innate immune system to the tumor site, promote antigen spread, and overcome resistance mechanisms like T-cell exhaustion or antigen escape, thereby broadening the anti-tumor response initiated by standard TCE therapies.
