TCIP1 is a first-in-class transcriptional/epigenetic chemical inducer of proximity (TCIP) designed for the treatment of B-cell malignancies, specifically diffuse large B-cell lymphoma (DLBCL). It is a bifunctional small molecule that recruits the transcriptional coactivator BRD4 to genomic sites bound by B-cell lymphoma 6 (BCL6). This recruitment effectively "rewires" the epigenetic state of the cell by converting BCL6 from its native role as a transcriptional repressor into a transcriptional activator. This functional flip induces the expression of BCL6 target genes that are typically silenced in lymphoma cells, leading to tumor cell death. Developed through research at Stanford University and MD Anderson Cancer Center, TCIP1 has demonstrated significant efficacy in BCL6-high patient-derived xenograft (PDX) models and has shown potential in overcoming resistance driven by epigenetic plasticity when used in combination with lenalidomide.
