TCP-25 is a synthetic peptide drug based on the 25 C-terminal amino acids of human thrombin, with a molecular weight of approximately 3088.6 Da. It is formulated as a hydrogel for topical application and is under development primarily for the treatment of epidermolysis bullosa (EB), a group of rare genetic disorders characterized by fragile, blistering skin and chronic wounds. TCP-25 acts as an antimicrobial peptide with dual action—directly targeting infection by interacting with bacterial lipopolysaccharide and other bacterial products, while also modulating excessive inflammation through interaction with the cell receptor CD14. This mechanism enables both antimicrobial effects (including activity against multidrug-resistant bacteria) and anti-inflammatory effects that promote wound healing. The drug has shown positive safety, tolerability, pharmacokinetic data (with no systemic uptake), and signs of wound healing efficacy in early clinical studies[2][3][4][5].
