TCRβ-directed CAR-T cells are an investigational cell therapy designed to treat T-cell malignancies, such as T-cell leukemia and T-cell lymphoma. This approach involves engineering T cells to express a chimeric antigen receptor (CAR) that specifically targets the T-cell receptor beta (TCRβ) chain. A key strategy in this field involves targeting one of the two mutually exclusive TCRβ constant regions (TRBC1 or TRBC2). Because a single T-cell clone (including malignant ones) expresses only one of these regions, targeting the specific TRBC involved in the malignancy can eliminate the cancer cells while preserving the healthy T-cell population that expresses the alternative TRBC, thereby reducing the risk of total T-cell aplasia and severe immunosuppression. This specific program is being evaluated in Phase 1 clinical trials by Xiamen University.
