TCR-T104 is an adoptive cell therapy consisting of T cells engineered to express a specific T-cell receptor (TCR), designated T104, which targets the KRAS G12V neoantigen. KRAS G12V is a prevalent driver mutation in several solid tumors, including colorectal, lung, and pancreatic cancers. The therapy is designed to recognize the KRAS G12V peptide presented by human leukocyte antigen (HLA) class I molecules on the surface of malignant cells, triggering T-cell activation and tumor cell lysis. Research suggests that the efficacy of TCR-T104 is significantly enhanced when combined with lymphodepleting chemotherapy, which upregulates immunoproteasome activity and HLA-I expression, thereby increasing the presentation of the target neoantigen.
