Overview

TCR-transduced peripheral blood lymphocytes are autologous human peripheral blood lymphocytes (PBLs) that have been genetically modified to express a specific tumor antigen-reactive T cell receptor (TCR). This is achieved by isolating the genes encoding an αβ T cell receptor from a tumor antigen-specific clone and introducing them into patient-derived PBLs using viral vectors such as retrovirus or lentivirus. The resulting engineered cells can recognize and target cancer cells presenting the relevant peptide-MHC complex. This approach is a form of adoptive cell therapy and is being investigated primarily for solid tumors such as melanoma. The mechanism of action involves redirecting the specificity of the patient's own immune cells to recognize and kill tumor cells expressing specific antigens[1][2][3][5][6].
