TCR-TREGs (T cell receptor-engineered regulatory T cells) are a form of cellular immunotherapy that combines the immunosuppressive functions of regulatory T cells (Tregs) with T cell receptor (TCR) engineering technology. This approach allows for the creation of Tregs with specific antigen recognition capabilities, enabling targeted immunosuppression. Tregs naturally maintain immune homeostasis and prevent autoimmunity through various suppressive mechanisms, including contact-dependent methods (direct cytolysis, dendritic cell disruption) and contact-independent methods (suppressive cytokine release, metabolic dysregulation). By engineering these cells with specific TCRs, TCR-TREGs can be directed to recognize particular antigens, potentially allowing for more precise control of immune responses in conditions requiring immunological tolerance.
