Overview

TCR1188-ABC cells are an autologous, base-edited T-cell receptor (TCR) engineered T-cell therapy developed by the University of Pennsylvania. The therapy consists of CD4+ and CD8+ T cells modified to express a TCR (TCR1188) specific for the KRAS G12V mutation, alongside a single-chain variable fragment (scFv) targeting the inhibitory receptor ILT4 (LILRB2). By targeting KRAS G12V, the cells are directed to recognize and eliminate tumor cells harboring this specific mutation, while the anti-ILT4 component is designed to block immunosuppressive signaling within the tumor microenvironment, potentially enhancing T-cell persistence and activity. It is currently being investigated in a Phase I trial for advanced solid tumors, including pancreatic, colorectal, and non-small cell lung cancers.
