Overview

TCRT-ESO-A2 is an autologous, affinity-enhanced T cell receptor (TCR)-engineered T cell therapy designed to target the tumor-associated antigen NY-ESO-1 in HLA-A*02:01 positive patients with advanced solid tumors. The therapy involves collecting a patient's own (autologous) T cells, stimulating and genetically modifying them ex vivo using a lentiviral vector to express a high-affinity, NY‑ESO‑1-specific TCR, and then reinfusing these engineered cells back into the patient. The goal is for these modified cells to recognize and attack cancer cells expressing NY‑ESO‑1. It was initially developed by Axis Therapeutics Limited—a joint venture between Athenex and Xiangxue Life Sciences—with clinical development also involving PharmaEssentia in Taiwan. The primary indication has been advanced solid tumors that are positive for NY‑ESO‑1[3][5][6].
