TCX-101 BRA#3 is an experimental T-cell receptor (TCR) engineered T-cell therapy being developed by BlueSphere Bio for the treatment of hematologic malignancies. The therapy consists of autologous or allogeneic T cells genetically modified to express a specific TCR that targets cells presenting 'Blood Restricted Antigen #3' (BRA#3). These blood-restricted antigens are expressed exclusively within the hematopoietic system, which allows the therapy to selectively eliminate malignant blood cells while avoiding off-target toxicity in non-hematopoietic tissues like the lungs, liver, or brain. This lineage-specific targeting approach is designed to provide a safer and more effective treatment option for patients with leukemias and other blood cancers who may not have suitable donors for stem cell transplants or who have relapsed after standard therapies. As of 2024, the candidate is in preclinical development.
