Overview

TCX-101 BRA#4 is a preclinical T-cell receptor (TCR) engineered T-cell therapy developed by BlueSphere Bio. It is a component of the TCX-101 clinical program, which focuses on targeting minor histocompatibility antigens (miHAs) that are exclusively expressed on hematopoietic cells. This specific candidate is engineered to target Blood Restricted Antigen #4 (BRA#4). The therapy is designed for use in patients with high-risk hematologic malignancies, such as acute myeloid leukemia (AML), acute lymphocytic leukemia (ALL), and myelodysplastic syndrome (MDS), particularly following allogeneic hematopoietic stem cell transplantation (alloHSCT). By targeting antigens restricted to the blood, the therapy aims to provide a potent graft-versus-leukemia (GVL) effect while minimizing the risk of graft-versus-host disease (GVHD) in non-hematopoietic tissues. The TCR was identified using BlueSphere's proprietary TCXpress platform.
