TCX-101-MMAE is a preclinical-stage antibody-drug conjugate (ADC) developed by Tacalyx GmbH for the treatment of solid tumors. It comprises a novel monoclonal antibody, TCX-101, which targets a specific tumor-associated carbohydrate antigen (TACA) overexpressed on various cancer cells, including gastrointestinal, lung, and breast cancers. The antibody is conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) with a drug-to-antibody ratio (DAR) of 4. TCX-101-MMAE is designed to internalize upon binding to TACAs on the cell surface, releasing MMAE to induce cell cycle arrest and apoptosis. Preclinical studies have demonstrated significant tumor growth inhibition in melanoma and breast cancer xenograft models with a favorable safety profile.
