TCX-201 DXd

TCX-201 DXd is a novel antibody-drug conjugate (ADC) developed by Tacalyx GmbH, currently in preclinical development for the treatment of various solid tumors. It consists of TCX-201, a high-affinity, fully humanized monoclonal antibody targeting a specific tumor-associated carbohydrate antigen (TACA) that is overexpressed in gastrointestinal cancers, pancreatic cancer, colorectal cancer, and non-small cell lung cancer (NSCLC). The antibody is conjugated to Deruxtecan (DXd), a potent topoisomerase I inhibitor, via a cleavable linker with a drug-to-antibody ratio (DAR) of 8. Preclinical studies have demonstrated that TCX-201 DXd exhibits superior internalization and specificity, leading to effective tumor cell killing in both cell-line-derived and patient-derived xenograft models.
