TD-1058

TD-1058 is an investigational small molecule inhibitor of ALK5, the kinase domain of the Transforming Growth Factor-β (TGF-β) type 1 receptor. It is being developed as an inhaled therapy for idiopathic pulmonary fibrosis (IPF), a progressive and fatal lung disease characterized by chronic inflammation and fibrosis. By inhibiting the TGF-β pathway, which drives fibroblast to myofibroblast transition—a key process in IPF pathology—TD-1058 aims to reduce fibrotic progression in lung tissue. Preclinical and early clinical data indicate that inhaled TD-1058 leads to dose-dependent reductions in TGF-β-induced signaling markers in bronchoalveolar cells[1][2][5].
