Overview
TDENV-PIV with AS03B adjuvant is an investigational tetravalent dengue virus purified inactivated vaccine (PIV) formulated to protect against all four serotypes of the dengue virus (DENV-1–4). The vaccine consists of non-attenuated viruses for each serotype, propagated in Vero cells and inactivated with formalin. It is combined with the AS03B adjuvant system—an oil-in-water emulsion containing DL-α-tocopherol and squalene—to enhance immunogenicity. Clinical studies have shown that this combination elicits robust neutralizing antibody responses across all four DENV serotypes and has an acceptable safety profile. The primary indication is prevention of dengue infection[2][5][8]. The vaccine was developed by the Walter Reed Army Institute of Research (WRAIR), often in collaboration with GlaxoSmithKline.
