TDI01

TDI01 is an innovative, oral, highly selective small molecule inhibitor of Rho-associated protein kinase 2 (ROCK2), developed by Beijing Tide Pharmaceutical (a subsidiary of Sino Biopharmaceutical Limited) and Graviton Bioscience. It is the first ROCK2 inhibitor domestically developed in China. The drug acts by inhibiting ROCK2, which leads to suppression of pro-inflammatory Th17 cells, promotion of regulatory T cells, restoration of immune homeostasis, and direct anti-fibrotic effects through inhibition of fibroblast differentiation and induction of myofibroblast apoptosis. This dual mechanism underlies its potential in treating chronic graft-versus-host disease (cGVHD), idiopathic pulmonary fibrosis (IPF), pneumoconiosis, liver fibrosis, COVID-19-related acute lung injury/acute respiratory distress syndrome (ALI/ARDS), and other fibrotic or inflammatory diseases. Preclinical studies have shown significant efficacy in models for inflammation and fibrosis; clinical trials have demonstrated good safety profiles[1][3][4][5][6][9][10].
