TDM-601 is a small interfering RNA (siRNA) therapeutic designed to silence the expression of Ribophorin II (RPN2). RPN2 is a subunit of the N-oligosaccharyltransferase (OST) complex, which is responsible for the N-glycosylation of proteins in the endoplasmic reticulum. In many solid tumors, particularly breast and esophageal cancers, RPN2 is overexpressed and plays a critical role in the glycosylation and stabilization of P-glycoprotein (MDR1), a well-known multi-drug resistance factor. By silencing RPN2, TDM-601 reduces the levels of functional P-glycoprotein, thereby reversing resistance to chemotherapeutic agents such as docetaxel. Additionally, RPN2 silencing has been shown to inhibit tumor growth and metastasis by affecting other glycosylated oncogenic proteins. Developed by Toray Industries, TDM-601 has been evaluated in Phase 1 clinical trials for the treatment of HER2-negative breast cancer and other solid tumors.
