TDM-812 is an investigational RNA interference (RNAi) therapeutic developed by 3-D Matrix, Ltd. for the treatment of refractory breast cancer. It consists of a small interfering RNA (siRNA) targeting **Ribophorin II (RPN2)**, formulated within a self-assembling peptide hydrogel (0.5% AGK/glucose). RPN2 is a key component of the oligosaccharyltransferase complex that facilitates the N-glycosylation of P-glycoprotein; its overexpression is linked to multidrug resistance and poor clinical outcomes in breast cancer patients. By silencing RPN2, TDM-812 aims to reduce the stability and activity of P-glycoprotein, thereby restoring the sensitivity of cancer cells to chemotherapeutic agents like anthracyclines and taxanes. The drug is designed for weekly **intratumoral** administration, utilizing the hydrogel platform to provide localized delivery and sustained release of the siRNA within the tumor.
