Drug intelligence / Profile preview
TDP-43 oligomer-specific monoclonal antibody (including the lead clone TDP-O9) is a conformational monoclonal antibody developed by Academia Sinica. It specifically targets misfolded TAR DNA-binding protein 43 (TDP-43) oligomers, which are neurotoxic aggregates implicated in the pathogenesis of several neurodegenerative diseases, including amyotrophic lateral sclerosis (ALS), frontotemporal dementia (FTD), and Alzheimer's disease (AD). Unlike standard TDP-43 antibodies that target monomeric or physiological forms of the protein, this antibody selectively binds to pathological TDP-43 oligomers without perturbing native TDP-43. In preclinical models of ALS, intravenous administration of the antibody has been shown to improve motor function, increase motor neuron survival, reduce neuroinflammation, and promote neuromuscular junction reinnervation. The antibody is currently in preclinical development, with humanization completed.
