Overview

TDP-747 is a boron-containing amino acid analogue designed for boron neutron capture therapy (BNCT) to treat refractory cancers. It is selectively taken up by cancer cells via the large neutral amino acid transporter 1 (LAT1), which is upregulated in many tumors, delivering boron-10 to tumor sites. Upon neutron irradiation, the captured neutrons trigger a localized nuclear reaction that destroys cancer cells while sparing healthy tissue. In preclinical studies, TDP-747 demonstrates a superior tumor-to-healthy cell boron ratio of 13:1 compared to the standard agent BPA's 3:1 ratio, potentially improving BNCT efficacy for indications such as advanced head and neck cancer, gliomas, and melanomas. Developed by Taeyi Life Sciences (TLS), it is part of their pipeline to enhance targeted radiotherapy for hard-to-treat cancers.
